Stroll jumped from fourth to second off the line and was quickly all over race leader Joel Eriksson, who locked up at the hairpin.
Despite being forced to the outside at the Mercedes turn, Stroll made a move to edge past the Swede but lost momentum on the kerbs on the exit as Eriksson retook the lead.
There was no stopping the reigning champion next time around, though, as he went side-by-side with the Motopark driver at the hairpin and went past him at the following corner.
Stroll then cruised to the finish to win from Eriksson and score his first hat-trick of the season and his 14th victory.
Sergio Sette Camara spent the first half of the race in fourth but he was under attack from Jake Hughes throughout - and the Carlin driver eventually made the move stick.
Hughes then went after Nick Cassidy and arrived to the back of the Kiwi driver with three minutes remaining.
The GP3 regular took advantage of a small lock-up at the Mercedes turn to overtake Cassidy and claim a podium finish in his debut F3 round.
Hitech's Alexander Sims passed Sette Camara for fifth, the Brazilian settling for sixth.
Ben Barnicoat scored six points in seventh, followed by Prema duo Ralf Aron and Maximilian Gunther.
VAR driver Anthoine Hubert took the final point-scoring position in 10th.
